Description A rapid and sensitive method has been developed for the estimation of intracellular lipids accumulated in oleaginous yeast cells [Rhodotorula gracilis CFR 1 used for present studies]. The cells are stained with Sudan Black B and the absorbance is measured at 580 nm using unstained cells as control. A linear relationship is observed between absorbance and lipid concn. in fermentation broth. The values predicted using this relationship were compared with those obtained using the conventional method of solvent extraction followed by gravimetric estimation and a close agreement has been obtained.
