A new series 2-(1<i>H</i>-benzimidazol-2-yl)-6-substitutedthieno[2,3-<i>b</i>]quinolines <b style="">4</b> have been synthesized by a one-step process in which 2-(chloromethyl)-1<i>H</i>-benzimidazole <b style="">2</b>/2-(mercaptomethyl)-1<i>H-</i>benzimidazoles <b style="">6</b> undergo nucleophilic substitution and cyclisation with 2-mercaptoquinoline-3-carbaldehyde <b style="">1</b>/2-chloroquinoline-3-carbaldehyde <b style="">5</b>.<b style=""> </b>The compounds have been synthesized by two methods to select a method of higher yield. The newly synthesized compounds have been characterized by IR, CHNS analysis, LC-MS and <sup>1</sup>H NMR and also screened for antibacterial activity.
